I had a green seedless grape growing on my arch for about 3 years, making green seedless grapes. I planted two muscadine grapes on the arch, and once they started flowering, my seedless grape started making seeds. The grape was a "free" plant from Raintree, listed as Einset, but turned out to be a green seedless, so who knows what it is. My muscadines are Carlos and Isons.

I have heard of this with other types of plants - so this is plausible. Your seedless grape wanted a pollinator for viable seeds, and you accidentally gave it one. Some seedless things will start setting seed with age or cold stress (Cottonwoods come to mind), but you are probably right about the Muscadines in this case.
If your grape was grafted and growth below the graft line happens you can often get another variety or grapes with seeds in them. I have bought many a grape that were supposedly seedless...that turned out to have seeds. I think in most cases I wasnt sold what I thought I was buying though I have heard that seedless varieties can become seeded for a number of reasons including stress.
